Z-Gly-Pro-OH is a Z (carbobenzyloxy)-protected dipeptide of glycine and proline, widely used as a building block in peptide synthesis. The Z group protects the N-terminal amino group of glycine, enabling selective activation and coupling of the free C-terminal carboxylic acid of proline to other amino acids or resins. This facilitates the incorporation of the Gly-Pro sequence into larger peptides and proteins, particularly useful for collagen-like structures where Gly-Pro-Pro repeats are common. It is also applied in solution-phase synthesis and biochemical research on enzyme mechanisms, protease substrates, and protein interactions. The Z protecting group provides excellent orthogonality, stability under basic/acidic conditions, and facile removal by catalytic hydrogenation.
