Used historically in cancer treatment, particularly for acute leukemias, due to its ability to inhibit dihydrofolate reductase, disrupting DNA synthesis in rapidly dividing cells. It was one of the first antifolate agents employed in chemotherapy. Also used in research settings to select cells in hybridoma technology and mutagenesis studies. Its use in clinical medicine has largely been replaced by safer derivatives like methotrexate.
