Acetyl tetrapeptide-15 is a peptide sequence derived from endomorphin-2 (Tyr-Pro-Phe-Phe-NH2), a human μ-opioid agonist. It has a skin desensitizing effect, thus reducing allergies and skin irritation. It is commonly used in products to reduce irritation, allergies and skin sensitivity.
Acetyl tetrapeptide-15 was developed with the aim of reducing skin hyperreactivity (decreased skin sensitivity) causing chronic inflammation and neuropathic pain through the activation of neurons in μ-opioid receptors.
The efficacy of this peptide has been demonstrated both in vitro and on real skin, with acetyl tetrapeptide-15 being tested for its ability to modulate CGRP release. CGRP is released following TRVP1 activation by Capsaisin.
The test was performed by incubating sensory neurons with acetyl tetrapeptide-15 (0.0003% and 0.001%), capsazepine (10 μM, a TRPV1 antagonist), or verapamil (100 μM, a calcium channel blocker) for 6 h and then exposing them to KCl and capsaicin. It was found that 0.001% acetyl tetrapeptide-15 significantly reduced CGRP release when neurons were exposed to both capsaicin and KCl, more effectively than 10 μM capsazepine.
A blinded clinical study demonstrated the ability of acetyl tetrapeptide-15 to reduce skin sensitivity following capsaicin exposure in 20 subjects. The protocol began by applying increasing concentrations of capsaicin solution intranasally to determine the concentration that produced discomfort. Then, a 0.0015% solution containing acetyl tetrapeptide-15 was applied twice daily for four days, and the application was repeated to increase the capsaicin concentration. Overall, there was a significant increase in the capsaicin threshold, demonstrating its efficacy.
